Understanding the Trypan Blue Dye Exclusion Method

The trypan blue dye exclusion technique differentiates live cells from dead ones based on membrane integrity. Live cells exclude the dye, appearing bright under microscopic analysis, while dead cells absorb the dye, appearing darker. The Vi-CELL® XR system captures images of cells as they pass through the measurement zone, using software algorithms to calculate the percentage of viable cells based on this contrast.

Streamlined Reagent Integration with Color-Coded System

The Vi-CELL® XR utilizes a reagent system comprising four color-coded lines for straightforward installation

  • Blue: Trypan Blue Stain

  • Green: Buffer Solution

  • Red: Disinfectant

  • Yellow: Cleaning Agent

Proper connection of these lines is vital; incorrect installation can lead to inaccurate data. The system includes LED indicators to confirm correct priming and functionality of each reagent line, as well as the status of the waste container.

These kits serve as direct replacements, containing all necessary components—including trypan blue, buffer, disinfectant, and cleaning solutions—packaged in color-coded bottles for easy identification and installation. Installation and Priming Instructions

To install the Cell Viability Reagent Pack:

  1. Place the reagent pack inside the Vi-CELL® XR reagent compartment.

  2. Connect each color-coded line to the corresponding port on the reagent pack.

  3. Access the instrument software toolbar, select “Instrument,” then “Replace Reagent Pack” to initiate the priming process.

  4. Verify that all five LED indicators are illuminated, confirming successful priming. If any indicators are not lit, select the “Prime” function again to complete the process.

Regular monitoring of the waste container’s LED indicator is recommended to prevent overfilling and ensure uninterrupted operation.
